Today, the Trump administration released a plan for how it plans to distribute a Covid-19 vaccine when one becomes available. The plan, which aims to begin next year, calls for giving the vaccine first to healthcare workers and people who are more at risk for severe illness. But others are saying that even a push to vaccinate at the beginning of next year may be optimistic. CDC Director Robert Redfield told a Senate Appropriations Subcommittee today that he believes a Covid-19 vaccine won’t be available for most Americans until late summer or early fall of 2021. Even if a vaccine is rolled out as soon as possible, it will still be a long time until things return to our pre-pandemic version of normal. Dr. Soumya Swaminathan, chief science officer at the World Health Organization, said this week that it will likely take until at least 2022 for enough people around the world to build immunity to Covid-19 through vaccination that we can resume our “normal life.”

Moderna vordert goed met zoektocht naar coronaproefpersonen

(ABM FN-Dow Jones) Moderna bereidt zich actief voor op een eventuele marktlancering van zijn coronavaccin. Dit meldde het Amerikaanse biotechbedrijf donderdag bij aanvang van zijn Research & Development Day.

Het biotechbedrijf zei dat tot nu toe 25.296 deelnemers zich hebben ingeschreven voor de Fase 3 COVE-studie met het coronavaccin. Er zijn in totaal 30.000 vrijwilligers voor het onderzoek nodig.

Inmiddels hebben 10.025 deelnemers hun tweede vaccinatie ontvangen.

Om extra transparant te zijn heeft Moderna het protocol van het onderzoek online beschikbaar gesteld.

Naast het Fase 3-onderzoek met het coronavaccin heeft het biotechbedrijf nog vier kandidaatmedicijnen in Fase 2 zitten.

The U.S. Patent Trial and Appeal Board ruled against Moderna in an intellectual property dispute with Arbutus Biopharma, stating Thursday that a key technology to deliver messenger RNA, or mRNA, was invented by the latter company. Moderna had argued that the technology was "unpatentable."
Moderna's experimental coronavirus vaccine uses lipid nanoparticles to deliver mRNA to human cells, along with other candidates in the biotech's pipeline. Should the vaccine get to market, Moderna could now owe Arbutus substantial royalties, which could limit its "pricing flexibility and margin profile vs other players" developing coronavirus vaccines, SVB Leerink analyst Mani Foroohar wrote.
Arbutus has licensed much of the lipid nanoparticle intellectual property to a spinoff called Genevant, of which it shares ownership with Roivant Sciences. Arbutus stated that should Moderna be forced to sign a licensing agreement with Genevant because of the PTAB ruling, it would receive 20% of sales royalties.

US biotech company Novavax has signed an agreement to carry out the industrial production of its vaccine for Europe in Spain, once it becomes available, with Spanish pharmaceutical Zendal (through its subsidiary Biofabri).

Specifically, the Galician-based firm will be responsible for the production of the vaccine designed by Maryland-based Novavax for the European Union. The clinical trials are currently in Phase 2 and Phase 3 is expected to begin at the end of the third quarter 2020.

The Spanish Agency of Medicines and Health Products (AEMPS), dependent on the Ministry of Health, has affirmed that this fact is an “important” boost for Spain’s pharmaceutical industry.

Novavax is establishing a network of agreements to be able to manufacture its vaccine in the event it obtains marketing authorisation.

This announcement comes on top of the one from Spain’s Ministry of Health in July, related to the Spanish pharmaceutical company Rovi. It said Rovi will provide filling and finishing capacity for vials of the US firm Moderna’s vaccine. The latter wants to supply markets outside the United States starting in 2021.

The vaccine maker Novavax said Thursday that it would begin the final stages of testing its coronavirus vaccine in the United Kingdom and that another large trial was scheduled to begin next month in the United States.

It is the fifth late-stage trial from a company supported by Operation Warp Speed, the federal effort to speed a coronavirus vaccine to market, and one of 11 worldwide to reach this pivotal stage. Novavax, a Maryland company that has never brought a vaccine to market, reached a $1.6 billion deal with the federal government in July to develop and manufacture its experimental vaccine, which has shown robust results in early clinical trials.

The new study, known as a Phase 3 trial, is expected to enroll up to 10,000 people in the United Kingdom. Half of the volunteers will receive two doses of the experimental vaccine, 21 days apart, and the others will receive a placebo.

Although Novavax is months behind the front-runners in the vaccine race, independent experts are excited about its vaccine because its early studies delivered particularly promising results. Monkeys that were vaccinated got strong protection against the coronavirus. And in early safety trials, published early this month in The New England Journal of Medicine, volunteers produced strikingly high levels of antibodies against the virus.

It is not possible to make a precise comparison between early clinical studies of different vaccines, but John Moore, a virologist at Weill Cornell Medicine, said that the antibodies from Novavax were markedly higher than any other vaccine with published results. “You just can’t explain that away,” he said.

The Phase 3 trials will determine if Novavax can live up to that promise. Although the British trial is smaller than those of other leading vaccines, the vaccine’s potency means that it might deliver clear results well before the trial is over, said Natalie Dean, a biostatistician at the University of Florida. Researchers analyze clinical trial data at predetermined points during the study.

“That’s one of the reasons why we have those types of analyses — to catch a home run if something is working exceptionally well,” she said.

(ABM FN-Dow Jones) Nu het coronavirus weer oplaait in grote delen van Europa, de Verenigde Staten het virus nauwelijks onder controle krijgen en de winter in aantocht is, voeren farmaceuten een race tegen de klok om een werkzaam vaccin te ontwikkelen.

Deze week voegden Johnson & Johnson en Novavax zich bij drie andere vaccinkandidaten die eerder al de laatste fase van klinische onderzoeken bereikten.

Het Amerikaanse Novavax zal in de komende vier tot zes weken 10.000 vrijwilligers tussen de 18 en 84 jaar werven in het Verenigd Koninkrijk en op zijn vroegst start in oktober ook een Fase 3 studie met 30.000 proefpersonen in de VS, mits Novavax daar groen licht krijgt van de toezichthouders.

Johnson & Johnson kondigde deze week een grote Fase 3 onderzoeksstudie met zijn vaccinkandidaat aan in meerdere landen, waaronder de VS, Brazilië en Zuid-Afrika. Het potentiële vaccin heeft een Nederlands tintje, want het wordt ontwikkeld door J&J's dochter Janssen Vaccines in Leiden. De eerste resultaten van de klinische studie met 60.000 mensen worden op zijn vroegst begin 2021 verwacht.

De Amerikaanse farmareus Pfizer begon in juli al aan Fase 3 onderzoek naar een kandidaatvaccin dat het samen met het Duitse BioNTech ontwikkelt en ook het mogelijke coronavaccin van het Amerikaanse Moderna ging in juli de laatste onderzoeksfase in.

Moderna hoopt de werving van 30.000 vrijwilligers nog deze maand af te ronden en CEO Stéphane Bancel denkt dat het vaccin mogelijk al in oktober, maar waarschijnlijk in november en op zijn laatst in december veilig en werkzaam zal worden bevonden. Pfizer en BioNTech hopen uiterlijk eind oktober uitsluitsel te geven of hun kandidaatvaccin werkzaam is.

AstraZeneca ontwikkelt samen met de Universiteit van Oxford een Covid-19 vaccin en startte in augustus de laatste onderzoeksfase. Het Brits-Zweedse farmabedrijf was tot nog toe het enige dat zijn klinische studie moest stilleggen nadat een deelnemer een ontsteking kreeg aan het ruggenmerg. Toch verwacht het bedrijf eind dit jaar of begin volgend jaar genoeg data te hebben verzameld om een goedkeuringsaanvraag in te dienen.

De bekende Amerikaanse viroloog Anthony Fauci verklaarde deze week tegen een Senaatscommissie dat hij erop rekent dat ergens in november of december duidelijk wordt of en welke van de vijf vaccinkandidaten werkzaam en veilig zijn.

Vaccin hard nodig om economieën aan te jagen

Wereldwijd zijn er inmiddels ruim 32 miljoen coronagevallen en het zal niet lang meer duren voor het aantal sterfgevallen door het virus de grens van 1 miljoen bereikt, volgens actuele Covid-19 data van Johns Hopkins.

De zorgelijke opwaartse besmettingstrend zal het economisch herstel ook geen goed doen. ING stelt dat de V waarover marktkenners praten dan niet zal staan voor een V-vormig herstel van de mondiale economie, maar eerder voor virus en vaccin. Econoom Carsten Brzeski raamt de krimp van de Amerikaanse economie dit jaar op meer dan 4 procent, terwijl de eurozone er zelfs 8 procent op achteruit gaat in 2020. China moet het doen met een zeer magere groei.

Na het V-vormige herstel dat deze zomer aanvankelijk leek te ontstaan, verliezen de meeste economieën nu weer stoom en hoewel voor 2021 een geleidelijk herstel wordt voorspeld, denkt Brzeski van ING niet dat dit een V-vormig herstel zal zijn.

Evan Brown, Hoofd Multi-Asset Strategie bij UBS Asset Management, denkt dat "de tijdslijn voor een betrouwbaar vaccin en veranderingen in de monetaire impuls dit vertragende herstel zowel kunnen bespoedigen als in gevaar brengen."

UBS rekent erop dat 1 tot 3 van de huidige vaccinkandidaten in het vierde kwartaal wordt goedgekeurd, eerst door de FDA en daarna ook door Europese toezichthouders. Medisch personeel en andere essentiële beroepen zullen vervolgens als eerste het vaccin ontvangen en vanaf medio 2021 vermoedelijk ook de rest van de samenleving.

Hoe dan ook zal er deze winter volgens de marktkenner nog geen breed beschikbaar vaccin zijn in de VS en Europa. Er heerst onduidelijkheid over hoe goed en hoe lang een vaccin bescherming biedt tegen het virus en ook de distributie speelt een rol in het herstel van de wereldeconomie.

De marktkenner van UBS waarschuwt voor een grillig herstel van de wereldeconomie, omdat opkomende economieën buiten China en Rusland waarschijnlijk achter zullen lopen op ontwikkelde economieën in het verkrijgen van het vaccin.

Betrouwbaar vaccin biedt nieuwe beleggingskansen

Ook de financiële markten snakken naar een vaccin. Volgens Jefferies zijn drie punten daarbij van belang: de werkzaamheid, de distributie en het vertrouwen in een vaccin.

"Een vaccin zal de markt de zekerheid bieden dat we op enig moment kunnen terugkeren naar [het nieuwe] normaal", aldus Jefferies.

Een goedgekeurd vaccin biedt beleggers nieuwe kansen. Zo denkt vermogensbeheerder DWS dat sectoren als "live entertainment, hotels en luxe-winkels – ook bedrijven met een lagere marktkapitalisatie – het goed zouden moeten doen als er eenmaal een vaccin is."

En marktstrateeg Evan Brown van UBS voorspelt dat de techdominatie bij een beschikbaar vaccin afneemt. "De goedkeuring van een covid-vaccin zal eindelijk een rem zetten op de dominantie van de mega-cap techbedrijven. Andere beursfondsen zullen weer in lift zitten", aldus Brown.

"Wat betreft Amerikaanse aandelen keert het fundamentele tij ten gunste van een bredere aandelenblootstelling ten koste van de tech-zware Nasdaq 100. De aankondiging van een vaccin zal deze trend waarschijnlijk versterken, omdat het de kans op winstherstel buiten de technologiesector en andere bedrijven die van de crisis hebben geprofiteerd, vergroot."

Barron's article this morning "10 Stocks to Buy as the Health-Care Revolution Races Ahead, According to Experts". In it Geoffrey Porges, a senior biopharma analyst at SVB Leerink says.

Porges: When you talk to vaccine experts, nearly 100% say they have the most confidence in traditional adjuvant protein subunit vaccines, because we know that technology works. [An adjuvant is a substance that boosts the patient’s immune response to the protein in the vaccine.] Among smaller vaccine developers that we’re most confident about, Novavax [NVAX] is in the lead. Sanofi [SNY] and GlaxoSmithKline [GSK] are behind them, but catching up quickly. They have taken a similar approach, using Glaxo’s adjuvant and Sanofi’s antigen [a substance that causes the immune system to produce antibodies against it]. I have confidence in both the safety and efficacy of those products. They’re probably not going to be available until the first part of next year.

The viral-vector vaccine programs [using a weakened virus to transport pieces of a pathogen into the body to stimulate an immune response] are limited by the immunogenicity of the vector [its ability to provoke a response]. The messenger RNA programs [which instruct the body’s cells to make proteins] seem pretty reactogenic [side effect–inducing]; 80% of people getting them get high fevers. I am somewhat concerned about their safety and tolerability in large populations.

[MRNA] is the most prominent mRNA company, and probably the most controversial. BioNTech [BNTX], CureVac, and Arcturus Therapeutics Holdings [ARCT] are also involved. BioNTech, which has partnered with Pfizer [PFE], recently announced they had taken their fifth Covid vaccine construct into the clinic. That is either very confidence-inspiring, or very concerning.

Johnson and Johnson VS Novavax

J&J currently has one advantage over NVAX, as lance recently pointed out. They have one dose compared to two. Some other differences and similarities to consider:

Both refrigerate at room temp.

J&J has a goal of 1 billion doses per year and NVAX has over 2 billion however, since J&J only requires a single dose, it’s neck and neck with NVAX in terms of doses. (NVAX I’ve seen articles saying 2-3b)

NVAX plus
J&J is a live adenovirus and may struggle with recruitment and side effects.

NVAX is a protein with a good safety profile.

J&J I believe didn’t have an effective vaccine in either the older or younger population (someone have this?) I believe it didn’t work well for one population.

NVAX is one if not the first to test 65 and up and hiv patients.

NVAX ph3 in uk bc of its uniqueness, could gain approval in Europe before American fda, which will lift share price tremendously, giving the credibility behind a product.

NVAX has contracts in place to sell almost all of their doses already.

Final thoughts: J&J has the brand name and trump has been promoting them consistently. I don’t know their results timeline given present fda updates to the EUA guidelines however, my thought is NVAX wisely bought us insurance with the UK ph3 trial. If EUA is given to any other company, it should coincide with our Ph 3 results in UK.

Money seems safe here for the time being and I’m sure there will be breaking news of global covid surges in the coming weeks as well as some vaccines failing and giving dollars to those left standing.

Mee eens. Daarbij is er genoeg ruimte voor 2 succesvolle producenten en meer ook nog!

Wanneer in te stappen / bij te kopen? De koers is op de muziek vooruit gelopen, en elke kleine tegenvaller kan behoorlijke dalingen veroorzaken.

Wordt het echter een marktproduct dan kan ie nog maal 3 gaan...

Moderna boekt progressie in ontwikkeling coronavaccin

(ABM FN-Dow Jones) Moderna heeft vooruitgang geboekt in de ontwikkeling van een vaccin tegen het coronavirus. Dat meldde het Amerikaanse biotechbedrijf vannacht.

Het biotechbedrijf heeft in dit ontwikkelingsproces twee in leeftijd verschillende testgroepen op twee verschillende momenten twee in omvang verschillende doses toegediend die bij beide groepen in algemene zin goed werden getolereerd. In totaal zijn 40 volwassenen getest.

Deze eerste fase van de studie toont volgens Moderna aan dat het kandidaatvaccin antilichamen kan neutraliseren bij zowel oudere als jongere volwassenen.

De resultaten werden gepubliceerd in de The New England Journal of Medicine.

Ik vond het zeer de moeite waard.

MRNA results/comments
NEJM publication by MRNA showed a few things to discuss. First the study is small with ten pts per group. These groups are not actually representative of the elderly. The 50-65 averaged low 60s. The greater than 70 was 71 plus or minus 1.2 years. In other words the groups were 71 and had zero pts older than 74.
Results. Lots discussion of binding antibodies with titers in the millions. So what ? Same trick different day. The functional antibody neutralizing antibody titers are meager at 300 (70 year olds) 400 for the 50-65 group. It’s being touted by saying it’s almost as good for elderly as it was almost as good as it is for healthy adults. Yes. It’s almost as good as the weak response we saw earlier. Some day I hope they actually test their vaccine in an actual elderly population.

They loaded the age groups and got ok data. It is what it is, it’s meh.

Omdat Novavax toch wat achter ligt op de tijdlijn denk ik dat er nog een behoorlijke koopkans komt omdat de concurrentie de markt eerder bereikt. Dan zal het tegenvallen en kan Novavax scoren. Ik denk en wens dat het zo gaat lopen. Jij?
